Die AP-2a-
und AP-2b-Knockout-Studien
deuten darauf hin, daß jedes AP-2-Protein eine individuelle Funktion
in der Mausentwicklung besitzt. AP-2g
wird während der frühen Entwicklung extraembryonal in den Trophoblastenzellen
und während der späten Entwicklung in der Neuralleiste exprimiert.
Im Mittelpunkt dieser Arbeit stand die Frage nach der Funktion von AP-2gwährend
der Entwicklung der Maus. Nach der Restriktionskartierung und partiellen
Sequenzierung des genomischen Lokus klonierte ich einen Gen-Targetingvektor
und inaktivierte das AP-2g-Gen
durch homologe Rekombination in embryonalen Stammzellen. Durch Injektion
der ES-Zellen in Blastozysten, welche in die Uteri scheinschwangerer Ammenmäuse
transferiert wurden, konnte ich AP-2g-defiziente
Mäuse erzeugen und analysieren. Ich konnte zeigen, daß AP-2g
für die frühe Embryonalentwicklung essentiell ist, da die Nullmutanten
ab Tag 7,5 der Embryonalentwicklung im Wachstum retardieren und bis Tag
9,5 dpc. resorbiert werden. Meine Ergebnisse belegen, daß der Verlust
von AP-2g
zu einer verminderten Proliferation der Zellen im extraembryonalen Ektoderm
und im ektoplazentalen Konus führt, und die Anzahl von Riesenzellen
vermindert ist. Als Folge davon kann sich das aus Chorion und Allantois
gebildete Labyrinth nicht entwickeln, der Embryo wird nicht genügend
mit Nährstoffen versorgt und stirbt.
Experimente
an kultivierten Blastozysten zeigen, daß die Expression von Adenosindeaminase
(ADA) bei AP-2g-defizienten
Blastozysten stark verringert ist. ADA katalysiert als zentrales Enzym
des Purinstoffwechsels den Abbau des Zellgifts Desoxyadenosin. Wird zu
wenig ADA exprimiert, dann akkumuliert Desoxyadenosin, und die Zellen sterben
ab. Dies ist eine weitere mögliche Erklärung für den Tod
der Nullmutante. Meine Ergebnisse zeigen, daß AP-2g
eine essentielle Funktion bei der Entwicklung der extraembryonalen Gewebe
besitzt. AP-2g
scheint sowohl die Proliferation der Zellen im ektoplazentalen Konus, als
auch die Expression mindestens eines Zielgens, nämlich ADA, zu fördern.
Um den früh letalen Defekt im Trophoblasten der AP-2g-defizienten
Maus zu umgehen, erzeugte ich unter Verwendung des Cre/loxP-Rekombinasesystems
eine konditional AP-2g-defiziente
Mausline, die in eine Mox2Cre-Mausline eingekreuzt wurde. Der Mox2-Promotor
vermittelt die strikt epiblastspezifische Inaktivierung des AP-2g-Gens,
weshalb AP-2g
im Trophoblasten aktiv bleibt und die Analyse der AP-2g-Funktion
im Embryo selbst ermöglicht. Weiterhin wurde die konditionale AP-2g-Linie
in eine Wnt1-Cre-Mauslinie eigekreuzt, um die Aufgabe von AP-2g
bei der Bildung der Neuralleiste und die Funktion in den Neuralleistenderivaten
zu studieren.

Deficient and Conditionally Deficient Mice Lines - The role of AP-2g
during mouse development

Studies
of AP-2?
and AP-2?
knockouts indicate that both proteins have distinct functional roles during
murine development. AP-2?
is expressed in trophoblast cells during early embryogenesis and in the
neural crest during later developmental stages. In this work I addressed
the question of the role of AP-2g
using gene-knockout technologies. Therefore, after restriction mapping
and partial sequencing of the genomic locus I generated a gene targeting
vectorto inactivate the AP-2?
gene by homologous recombination in embryonic stem cells. The ES-cells
were injected in blastocysts and the latter was transferred into the uteri
of pseudopregnant foster mice. The germline transmitting offspring was
bred in order to generate and analyze mice deficient for AP-2g.
I
was able to demonstrate that AP-2?
is essential for early embryogenesis, for the nullmutant displays a significant
retardation in growth by day 7.5 dpc. and is resorbed by day 9.5 dpc..
The loss of AP-2?
leads to a reduction in cell proliferation in the extraembryonic ectoderm
and in the ectoplacental cone, as well to a reduced number of giant cells.
As a consequence the labyrinthlayer, derived from chorion and allantois
fails to form. The embryo suffers from deprivation of nutrients and, as
a consequence, dies.
In
addition, in vitro experiments with cultured blastocysts show that
the expression of adenosine deaminase (ADA) is highly reduced. ADA is a
purine catabolic enzyme that converts cytotoxic deoxyadenosine to deoxyinosine.
The reduced ADA expression leads to the accumulation of the toxin, and
thus to cell death. This could be a further explanation for the death of
the nullmutant.
My
results show, that AP-2g
plays an essential role in the development of extra-embryonic tissues.
AP-2g
seems to regulate the proliferation of cells in the ectoplacental cone
and the expression of at least one target gene, which is ADA.
By
using the Cre/loxP recombination system I generated a conditional AP-2?-knockout-mouse
to circumvent the early lethal defect in the trophoblast. This
mouse has been crossed with a Mox2Cre-mouse, because the Mox2-promoter
mediates a strict epiblast inactivation of the AP-2?-gene. Thus, AP-2?
is still active in the trophoblast and allows the analysis of AP-2? function
in the embryo proper. Furthermore,
the conditional AP-2?-mouse
has been crossed with a Wnt1-Cre-mouse to analyze the function of AP-2?
in the development of the neural crest and its derivatives.
