Zusammenfassung
Die Transduktion eines
extrazellulären Signals von der Plasmamembran zu selektiven Kompartimenten im
Inneren der Zelle erfordert die Bildung von Multiproteinkomplexen spezifischer
Zusammensetzung und Lokalisation. Adapterproteine, wie die
LIM-Domänen-Proteine, spielen eine wichtige Rolle bei der Rekrutierung von
Proteinen zu ihren Interaktionspartnern im Proteinnetzwerk. Das intrazelluläre
Adhäsionsprotein Trip6 und die kleinere, kernspezifische Isoform nTrip6 sind
beides LIM-Domänen-Proteine, die durch das trip6
Gen kodiert werden, jedoch in unterschiedlichen Zellkompartimenten lokalisiert
sind. Trip6 reguliert die Bildung von fokalen Adhäsionskomplexen an der
Plasmamembran, wohingegen nTrip6 im Nukleus als Koaktivator für die
Transkriptionsfaktoren AP-1 und NF-B fungiert. Die spezifische intrazelluläre
Verteilung von Trip6 und nTrip6 deutet daraufhin, dass die trip6 Protein-Isoformen unterschiedliche funktionelle Eigenschaften
der Zelle regulieren. Trip6 könnte die Zellmigration und Adhäsion und nTrip6
die Zellproliferation und Differenzierung modulieren. Jegliche Störung des
Gleichgewichts der Funktionen von Trip6 und nTrip6 könnte somit signifikante
Auswirkungen auf pathologische Prozesse, wie Entzündungsreaktionen, Tumorgenese
oder Metastasierung, zur Folge haben.
Bisher ist weder der Mechanismus der zur
Bildung der trip6 Isoformen führt,
noch die in vivo Relevanz der Trip6 und nTrip6 Funktionen bekannt. Ein Ziel
dieser Arbeit war daher eine konditionell trip6
defiziente Mauslinie zu generieren, um die Funktionen von trip6 in vivo zu
untersuchen. Anhand der konditionellen Gen-targeting Methode habe ich das trip6 Gen in murinen embryonalen
Stammzellen modifiziert. In diesen Zellen wurde das trip6 Gen durch Cre-DNA-Rekombinase Erkennungssequenzen
(LoxP-Sequenzen) flankiert. Die Cre-Expression führt zur Exzision von
LoxP-Sequenzen flankierten DNA-Abschnitten. Es ist jetzt möglich diese
embryonalen Stammzellen zu nutzen, um eine konditionell trip6 defiziente Mauslinie zu generieren, in der beide Trip6
Isoformen deletiert sein werden. Eine Strategie die Funktionen der zwei Trip6
Isoformen in vivo unterscheiden zu
können, wäre die Rekonstitution einer trip6
defizienten Maus mit nur einer der beiden Isoformen. Die Vorraussetzung dafür
war daher die Identifizierung des Mechanismus der zur Bildung von nTrip6
führt.
In dieser Arbeit ist es mir gelungen zu
zeigen, dass nTrip6 nicht durch Translation einer alternativ gespleißten trip6 mRNA, sondern durch alternative
Translationsinitiation an einem sekundären Startkodon gebildet wird. Sowohl in
Mutagenese-Experimenten als auch durch Verwendung spezifischer Morpholinos zur
sterischen Blockierung der Translationsinitiation, konnte ich zeigen, dass das
AUG-Kodon an Position 480 der trip6
mRNA für die Translation von murinem nTrip6 genutzt wird. Die alternative
Translationsinitiation an diesem Startkodon war mRNA-kappenabhängig und
benötigt keine internen ribosomalen Eingangsstellen (IRES). Des Weiteren konnte
ein Morpholino der gegen das sekundäre Startkodon gerichtet war und nur die
Translation von nTrip6 blockierte, die TPA-induzierte Aktivierung eines
AP-1-Zielpromotors inhibieren. Die selektive Manipulation der konventionellen
oder alternativen Translationsinitiation an der trip6 mRNA stellt jetzt eine Strategie dar, die Bildung und
Funktion von Trip6 oder nTrip6 in spezifischer Weise zu blockieren, um so deren
jeweilige Rolle bei der Regulation der Zellproliferation, Differenzierung und
Migration zu untersuchen. Darüber hinaus wird das Wissen über die Bildung der
beiden Trip6 Isoformen nützlich sein, ihre jeweiligen Funktionen in vivo zu untersuchen, indem eine trip6-defiziente Mauslinie entweder mit
Trip6 oder nTrip6 spezifisch rekonstituiert wird.

selective analysis of Trip6- and nTrip6-dependent functions in vivo
Abstract
Transduction of an extracellular signal from the plasma
membrane to specific compartments in the cell requires the formation of multi
protein complexes of distinct composition and localization. Adaptor proteins,
such as LIM domain proteins, play an important role in the recruitment of
proteins to their interaction partners within the protein network. The
intracellular adhesion protein Trip6 and its smaller, nuclear isoform nTrip6
are both LIM domain proteins encoded by the gene trip6 but are localized in different cellular compartments. Trip6
regulates the formation of focal adhesion plaques at the plasma membrane while
nTrip6 acts in the nucleus, as co-activator for the transcription factors AP-1
and NF-B.
Due to their specific intracellular distribution the two trip6 protein isoforms regulate different functional properties of
the cell. Trip6 might modulate cell migration and adhesion and nTrip6 might
regulate cell proliferation and differentiation. Any imbalance in Trip6 and
nTrip6 functions might have significant consequences in pathological situations
such as inflammation, cancer or metastasis.
Neither the mechanism by which the two isoforms are generated, nor the in vivo relevance of Trip6 and nTrip6
functions is known. Thus, one aim of this work was to generate a conditional trip6 deficient mouse strain to study
the functions of trip6 in vivo. I modified the trip6 gene by conditional gene targeting
in murine embryonic stem cells. In these cells the trip6 gene was flanked by Cre-DNA-recombinase recognition sites
(loxP-sites). Cre-expression leads to excision of loxP-sites flanked
DNA-sequences. It is now possible to use these embryonic stem cells to generate
a conditional trip6 deficient mouse line.
In this mouse, both Trip6 isoforms will be knocked out. To discriminate the
functions of the two isoforms in vivo,
one strategy would be to reconstitute the knockout mouse with only one isoform.
A prerequisite was thus to identify the mechanism of nTrip6 generation.
In this work, I showed that nTrip6 is not generated by translation of an
alternatively spliced trip6 mRNA but
through alternative translation initiation at a secondary start site. In
mutagenesis studies as well as by the use of specific morpholinos to sterically
block translation initiation, I showed that the AUG codon at position 480 in
the murine trip6 mRNA is utilized for
the translation of nTrip6. The alternative translation initiation at this site
was mRNA cap-dependent and did not require internal ribosome entry sites
(IRES). Furthermore a morpholino targeting the secondary AUG and knocking down
only nTrip6 repressed the TPA-induced activation of an AP-1 target promoter.
Thus the selective manipulation of conventional or alternative translation
initiation at the trip6 mRNA will now
serve as a strategy to specifically block the generation and function of Trip6
or nTrip6, in order to study their respective roles in cell proliferation,
differentiation and migration. Moreover, the knowledge about the generation of
the two isoforms will be valuable to study their respective functions in vivo, by the specific rescue of the
knockout mouse with either Trip6 or nTrip6.
